background: C9orf86 (Partner of ARF;PARF) binds p19(ARF), but not the p16(INK4A) isoform of CDKN2A. Knockdown of C9orf86 by RNA interference reduced the growth inhibitory activity of p19(ARF) and enhanced the rate of cell proliferation. Function: May enhance cellular proliferation. May reduce growth inhibitory activity of CDKN2A. Subcellular Location: Cytoplasmic and Nuclear. Note=Predominantly cytoplasmic. Proposed to shuttle between cytoplasm and nucleus. Post-translational modifications: Isoform 1 is O-glycosylated, while other isoforms are not. Similarity: Belongs to the small GTPase superfamily. Rab family. Protocol
The cells were fixed with 4% PFA (10min at room temperature)and then permeabilized with 90% ice-cold methanol for 20 min at-20℃. The cells were then incubated in 5%BSA to block non-specific protein-protein interactions for 30 min at room temperature .Cells stained with Primary Antibody for 30 min at room temperature. Acquisition of 20,000 events was performed.
